Description

Despite excellent initial responses to the powerful and specific new inhibitors of cancer driving mutations in non-small cell lung cancers like ALK or EGFR, patients inevitably experience recurrence of refractory disease- a process driven by Darwinian evolution. We seek to focus on the process of evolution itself as a targetable and rationally perturbable process. In this talk we will describe mathematical tools used to control large natural populations like those in game reserves or fisheries-termed evolutionary games- and how these can be applied to clinical oncology. We will then describe an in vitro assay we have developed to directly measure the parameters of these models for specific scenarios, and results of these measures for both ALK and EGFR mutated NSCLC. We will conclude by describing how to translate these findings and concepts to adaptive, evolutionary trial design.

1. Understand how tools from theoretical ecology can help us understand competition in heterogeneous tumors2. Understand how tumor ecology can allow resistance conferring mutations to persist in small numbers before therapy in the context of significant fitness costs3. Understand how an in vitro evolutionary game assay can describe intratumoral clonal competition.
