Description
This presentation will introduce spatial transcriptomic approaches for mapping the bone marrow microenvironment and revisit the historical concept of the erythroblastic island as the canonical niche for erythropoiesis. By comparing murine and human bone marrow architecture using an unbiased spatial transcriptomic approach, the talk will re-examine whether the classical macrophage-centered erythroblastic island model applies to human erythropoiesis and highlight distinct cellular interaction programs that support erythroid maturation in human bone marrow.

Objectives
1. The murine erythropoietic niche is organized as an erythroblastic island centered on a distinct C1q-expressing macrophage population.
2. The human erythropoietic niche does not require central macrophages, but instead relies on ICAM-4–mediated cell–cell interactions between erythroblasts.
3. In patients with myelodysplastic syndromes, anemia is associated with decreased ICAM-4 expression in the erythroblasts and reduced erythroblast–erythroblast interactions.
